Advancing 3D Hepatic Spheroid-Array Approach for Hepatic Clearance Assessment

A recent publication from Sai Life Sciences highlights our work on a three-dimensional hepatic spheroid-array system for the assessment of hepatic clearance of small molecules and beyond-rule-of-five (bRo5) compounds, including PROTACs.
The study demonstrates how the 3D hepatic spheroid-array system provides a more physiologically relevant approach to predicting hepatic clearance, enabling improved characterization of metabolically stable compounds that are often challenging to assess using conventional suspension hepatocyte models.
As part of our broader focus on New Approach Methodologies (NAMs), we continue to advance physiologically relevant strategies that help answer increasingly complex ADME questions and support informed decision-making in drug discovery and development.
